My first suspicion would be that another application is using the microphone. Another thing to check is that the microphone is actually switched on through the volume controls of your sound card. To check this, right click the speaker icon in your system tray and select "open volume controls." Make sure the microphone is not muted and that the volume is set appropriately. It make take some trial-and-error to get the settings just right.
